DNA-encoded Libraries (DEL)
A robust split-and-mix procedure was implemented to produce the Macrocyclic DNA-encoded libraries in a combinatorial fashion. Each DNA-Macrocycle Conjugate is available for two synthetic cycles of decoration as a starting building block with an open diamine (1 variant) or an amino acid (2 variants) function. Reductive amination and amide bond formation are two of the most common reactions used in the DEL synthesis cycles. The combinatorial power of this approach is theoretically limited only by the number of available building blocks: acids, aldehydes, and amines, and could potentially lead to a library of billions of compounds. In practice, a reasonable diversity can be achieved by using sets of 96 (1 well-plate) monofunctional building blocks of each compatible functionality, creatively selected based on their molecular and electronic properties, price considerations, and reactivity. Each diamino acid Macrocyclic Scaffold can provide 73,728 of unique DNA-encoded structures.
